焦化废水中酚降解菌及其降解基因的研究   总被引:5,自引:4,他引:1  
酚类化合物是焦化废水的主要污染物,微生物降解在废水处理中起着主要作用.为获得焦化废水活性污泥中主要降解菌,本研究通过富集与平板涂布对某焦化公司的2个活性污泥中的降解菌进行了分离鉴定.通过BOX-PCR和16S rDNA序列分析去除重复菌株后,共获得分属于20个属的28个种的28株细菌,它们主要为变形菌纲βγ亚群,其中4株菌可能是潜在的新种.间甲酚富集后筛选得到2株高效降解菌株Pseudomonas monteilii GCS-AE-J-1 和Pseudomonas plecoglossicida GCS-AN-J-3;前者在48 h内对791 mg/L间甲酚的降解率达到94.6%,而后者对763mg/L间甲酚的降解率也达到了92.2%.通过PCR从菌株GCS-AE-14、 GCS-AE-J-1、GCS-AN-J-3和GCS-AN-3得到了苯酚羟化酶基因序列.本研究所获得的降解菌新颖多样,在工业焦化废水的处理中可能起着重要作用,有进一步研究开发的价值.  相似文献

从处理印染废水的活性污泥中分离得到2株苯胺降解菌,从菌落、细胞形态、生理生化及16S rRNA基因扩增测序等方面对2株菌进行了鉴定,并比较分析2株菌在好氧与缺氧条件下的苯胺降解、偶氮染料脱色及苯胺脱氨氧化酶基因tdnQ和黄素还原酶基因(fre)的携带情况.结果表明,2株菌属于Pseudomonas属和Shewanella属,分别命名为Pseudomonas sp.AN30和Shewanella sp.DN425.AN30菌株在振荡好氧条件下72h内对250mg/L苯胺的降解率为96.1%,DN425菌株的降解率为13.8%;在静置缺氧条件下AN30菌株的苯胺降解率为39.6%,DN425菌株的降解率仅为8.6%.DN425菌株在静置缺氧条件下4h内可将初始浓度为50 mg/L的偶氮染料酸性大红彻底脱色,而AN30菌株对酸性大红不具有脱色能力.以总DNA为模板,分别用tdnQ基因和fre基因特异性引物进行扩增,2株菌均能扩增出大小分别为380bp和630bp左右的目标条带,显示2菌株均携带有苯胺脱氨氧化酶基因和黄素还原酶基因.  相似文献

The global extent of macroalgal forests is declining, greatly affecting marine biodiversity at broad scales through the effects macroalgae have on ecosystem processes, habitat provision, and food web support. Networks of marine protected areas comprise one potential tool that may safeguard gene flow among macroalgal populations in the face of increasing population fragmentation caused by pollution, habitat modification, climate change, algal harvesting, trophic cascades, and other anthropogenic stressors. Optimal design of protected area networks requires knowledge of effective dispersal distances for a range of macroalgae. We conducted a global meta‐analysis based on data in the published literature to determine the generality of relation between genetic differentiation and geographic distance among macroalgal populations. We also examined whether spatial genetic variation differed significantly with respect to higher taxon, life history, and habitat characteristics. We found clear evidence of population isolation by distance across a multitude of macroalgal species. Genetic and geographic distance were positively correlated across 49 studies; a modal distance of 50–100 km maintained FST < 0.2. This relation was consistent for all algal divisions, life cycles, habitats, and molecular marker classes investigated. Incorporating knowledge of the spatial scales of gene flow into the design of marine protected area networks will help moderate anthropogenic increases in population isolation and inbreeding and contribute to the resilience of macroalgal forests. • Pig feces is the predominant excrement produced by animal husbandry in China. • The PF, Pig-1-BacTaqMan, and Pig-2-BacTaqMan MST assays showed better performance. • The pig-specific MST assays can contribute to managing the pig fecal pollution. In China, pig feces is the predominant source of excrement produced by animal husbandry. Improper use or direct discharge of pig feces can result in contamination of natural water systems. Microbial source tracking (MST) technology can identify the sources of fecal pollution in environmental water, and contribute to the management of pig fecal pollution by local environmental protection agencies. However, the accuracy of such assays can be context-dependent, and they have not been comprehensively evaluated under Chinese conditions. We aimed to compare the performance of five previously reported pig-specific MST assays (PF, Pig-Bac1SYBR, Pig-Bac2SYBR, Pig-1-BacTaqMan, and Pig-2-BacTaqMan, which are based on Bacteroidales 16S rRNA gene markers) and apply them in two rivers of North China. We collected a total of 173 fecal samples from pigs, cows, goats, chickens, humans, and horses across China. The PF assay optimized in this study showed outstanding qualitative performance and achieved 100% specificity and sensitivity. However, the two SYBR green qPCR assays (Pig-Bac1SYBR and Pig-Bac2SYBR) cross-reacted with most non-pig fecal samples. In contrast, both the Pig-1-BacTaqMan and Pig-2-BacTaqMan assays gave 100% specificity and sensitivity. Of these, the Pig-2-BacTaqMan assay showed higher reproducibility. Our results regarding the specificity of these pig-specific MST assays differ from those reported in Thailand, Japan, and America. Using the PF and Pig-2-BacTaqMan assays, a field test comparing the levels of pig fecal pollution in rivers near a pig farm before and after comprehensive environmental pollution governance indicated that pig fecal pollution was effectively controlled at this location.  相似文献

Hexavalent chromium-tolerant (1500?mg/L) bacterium MW1 was isolated from harbour water of Paradip Port and evaluated for Cr(VI) reduction potential. The isolate was identified as Exiguobacterium indicum by biochemical and 16S rRNA gene sequence methods. Salt tolerance of the bacterium was evaluated in a wide range of NaCl concentrations (0.5–13%, w/v). The Cr(VI) reduction of the strain was evaluated and optimised with varied Cr(VI) concentrations (100–1000?mg/L), pH (5.0–9.0), temperature (30–40°C) and shaking velocity (100–150?rpm) in two different minimal media (M9 and Acetate). Under optimised conditions, after 192?h of incubation nearly 92%, 50% and 46% reduction in the M9 minimal medium and 91%, 47% and 40% reduction in the acetate minimal medium were observed for 100, 500 and 1000?mg/L of Cr(VI), respectively. The exponential rate equation for Cr(VI) reduction yielded higher rate constant value, that is, 1.27?×?10?2?h?1 (M9) and 1.17?×?10?2?h?1 (Acetate) in case of 100?mg/L and became lower for 500 and 1000?mg/L Cr(VI) concentrations. Further, the association of bacterial cells with reduced product was ascertained by Fourier transform infrared spectrometer, UV–Vis–DRS and field-emission scanning electron microscope–energy-dispersive X-ray analyses. The above study suggests that the higher reducing ability of the marine bacterium E. indicum MW1 will be suitable for Cr(VI) reduction from saline effluents.  相似文献
